AxolHumaniPSC-DerivedNeuralStemCells(NSCs)arederivedfromintegration-free,inducedpluripotentstemcells(iPSCs)underfullydefinedneuralinductionconditions.TheNSCsexpresstypicalMarkersofcerebralcorticalneuralstemandprogenitorcellssuchasPAX6,FOXG1andnestin,andspontaneouslyformpolarizedneuraltube-likerosettestructureswhenplatedasamonolayerinculture(seebelow).Additionally,AxolNSCsarecapableofgeneratingaspectrumofcerebralcorticalexcitatoryandinhibitoryneuronsthatareelectricallyactiveandhavetheABIlitytoformfunctionalsynapsesandcircuitsinvitro.Afterthawingandplating,theneuralstemcellsterminallydifferentiatetoacquirematureelectrophysiologicalproperties,andformfunctionalsynapticnetworksoveraperiodof40~50days.
